Glutamate is the neurotransmitter of most excitatory synapses in the mamalian central nervous system (CNS).Several glutamate analogues are selective group III receptor agonists. They differ from the other analogues by the presence of an additional distal acidic function that provides both higher affinity and selectivity. That function is either a third carboxylic group as in ACPT-I, (+)-ACPT-III,and (S)-3,4-DCPG or the second acidity of a phosphonate group as in L-AP4,L-SOP,(S)-PPG and (1S,2R)-APCPr. Among these, L-AP4 is the most potent group III mGluR agonist with a potency of about 10- to 30-fold that of glutamate.
